(Using RFLP Technique)

DNA typing (also called DNA profile or DNA Fingerprinting) is the process where the genomic DNA of an organism is analyzed to examine several specific, variable DNA sequences located throughout the genome. DNA Fingerprinting is used in forensic science (paternity and crime determination and also for the identification of human remains). The experiment is based on the difference in the distance between successive restriction enzyme sites due to mutation.

MEDOX-BIOTM Southern Blotting Teaching Kit

Southern blotting is the deduction of specific DNA sequence using a DNA probe of known sequence. The DNA is denatured neutralized and transferred from gel to nitrocellulose membrane by capillary action and baked at 80°C in vacuum oven for firm attachment. The blotted DNA can be further used for hybridization.

Restriction mapping is a process of obtaining structural information on a piece of DNA by the use of restriction enzymes. Restriction mapping involves digesting DNA with a series of restriction enzymes and then separating the resultant DNA fragments by agarose gel electrophoresis. The distance between restriction enzyme sites can be determined by the patterns of fragments that are produced by the restriction enzyme digestion. In this way, information about the structure of an unknown piece of DNA can be obtained

Restriction enzymes cleave DNA at discrete nucleotide sequences thereby cutting DNA fragments. However enzyme methylase covalently joins methyl groups to adenine or cytosine residues within specific target sequences. MEDOX-BIOTM Preparation and Deduction Of Dioxigenin Labelled Dna Probe Teaching Kit

A PCR-derived digoxigenin-labeled DNA probe was used for Epstein-Barr early RNA (EBER) in situ hybridization in formalin-fixed paraffin-embedded tissues. The results showed that the hybridization signal was morphologically distinct and the intensity of signal was comparable with those by RNA riboprobe. The advantages of using PCR-derived DNA probes for EBER in situ hybridization include:
(1) the synthesis of digoxigenin-labeled DNA probes is easy and simple by PCR; (2) the labeled amplification product can be used as a probe without further purification; (3) DNA probes are potentially more stable than RNA probes; and (4) the preparation of DNA probes is relatively efficient and rapid. It is concluded that this technique is an ideal candidate for detection of EBER expression in clinical specimens.

MEDOX-BIOTM Micrococcal Nuclease Digestion Of Purified Genomic DNA Teaching Kit

The basic unit chromatin is the nucleosome composed of DNA and histones. Each histone has an N-terminal tail that extends out side the DNA and these tail are the sites of numerous covalent modifications that contribute significantly to the regulation of chromatin structures.
Micrococcal nuclease (MNase) is an important enzyme in characterizing nucleosome structure across any specific gene in intact cells. MNase cleaves preferentially in the linker DNA that lies between individual nucleosomes. MNase cleavage can be used to map the boundaries of nucleosomes and determine their spacing.
