Agglutination reactions occur when antibodies (agglutinos) react with particulate antigens (agglutinogen) on a cell surface. The antigen-antibody reaction causes the cells to clump or agglutinate. When agglutination reactions involve the clumping of red blood cells, they are termed as heamagglutination.
ABO blood grouping was described by Karl Landsteiner. He reported 4 different types of human immunological blood groups. They are A, B, AB and O. These blood groups are genetically controlled. Type A blood has antigen A on the RBC. Type B has B antigens on the RBC. Type AB has both A and B antigens while O type blood has no antigens but only antibodies a and b. The AB antigens are inherited as two (one from each parent) of three alternative alleles are
dominant over O.

Widal test is a serological method used in the diagnosis of enteric fever, which is caused by the organisms of the genus Salmonella. They are Salmonella typhi, Salmonella paratyphiA,B and C. Enteric fevers due to S.typhi and S.paratyphi A,B and C are common in India . Antigens specifically prepared from this organism are used in the agglutination test to detect the presence of antibodies in patients sera, which are elucidated in response to infection by these bacteria.

Venereal diseases are sexually Transmitted diseases are transmitted by sexual intercourse. These include AIDS, SYPHILIS, GONORRHEA, nonspecific UTERITIS, etc., though there have been a number of diseases listed under VDRL (Venereal disease research laboratory). Our test is specific to syphilis.

Human diseases caused by Treponemes

3. Yaws - Treponema pallidum
4. Pinta - Treponema carateum
It is a flocculation test used to find out antibody against the reaginic antigens in syphilitic infection caused by T.pallidum. The presence of antibody in the patient's serum is detected by using a non specific antigen prepared from cariolipin, lecithin and cholesterol

Rheumatoid arthritis is a chronic inflammatory disease of the joints. The pathogenesis involves the formation of abnormal IgG, by the synovial lymphocytes. In response to the abnormal IgG, the immune system of the host produces IgM and IgG anti- immunoglobulin, which are called Rheumatoid factors (RF).
RA test antigen consists of polystyrene latex particles coated with specially purified human immunoglobulin. The suspension of coated latex particles agglutinate visibly when mixed with serum containing Rheumatoid factor in concentration equal to or greater than the sensitivity, by slide (qualitative) and (semiquantitative) methods respectively.

RPR antigen suspension is a carbon-containing antigen (cardiolipin), which detects regain antibody present in serum of syphilitic individuals. In a serum sample containing reagin antibody, flocculation occurs due to agglutination of the carbon particles of the RPR antigens, which appear as black clumps against the white background of the card. This co agglutination is read macroscopically. Non-reactive specimens show an even light grey colour.

Human chorionic gonadotropin (hCG) is a hormone produced by placenta very early in pregnancy. It is secreted in the urine approximately 20 days after the last menstrual period. The level increased rapidly till 60 - 80 days and then falls slightly from the peak and maintain the same level till 280 days
The test is based on antigen (b-hCG in urine sample) and antibody (anti b-hCG antigen) reaction. b-hCG, it present in urine sample neutralizes the anti b-hCG antibody/antiserum and hence does not allow agglutinating antigen (hCG antigen coated particulate antigen i.e. RBC) for a reactive sample. However, when b-hCG is absent in the urine sample, the free anti b-hCG antibody/antisera reacts with the hCG agglutinating antigen and results into agglutination.

Haemagglutination is based on the principle of agglutination reaction. All strains of New castle Disease (ND) virus agglutinate with chicken red blood cells. Neuraminidase viral proteins bind to the receptors on the membrane of red blood cells. This linking of red blood cells with viral particles, which result in clumping is what known as haemagglutination.
