Elimination ("curing") of a plasmid from a bacterial culture is the best test to substantiate the relation between a genetic trait and carriage of that specific plasmid by the culture. A phenotype linked to the presence of the plasmid will not be expressed in cured cultures. Reintroduction of the plasmid or a recombinant clone derivative carrying a specific genetic trait must be accompanied by regaining of the phenotype. Various protocols involving physical and chemical agents have been developed to cure plasmids. Depending on the nature of the bacterial host and /or the plasmid, some methods work better than others. Protocols for curing plasmids consist basically exposure of culture to different concentrations of the chemical agent or to higher than normal temperature, followed by plating and analysis of the presence of a trait carried by a plasmid. Colonies that lose the trait or then analyze for their plasmid content to confirm lose of the plasmid.

MEDOX-BIOTM Determination Of Tm Value Teaching Kit

The Tm value of a DNA molecule depends markedly on its base composition. DNA molecules rich in GC base pairs have a higher TM that those having an abundance of AT base pairs. For most of the DNA a sharp increase in O.D260 will be observed from 60OC to 80OC. The midpoint temperature of this increasing slope is called TM value or melting temperature of DNA.

Two types of methods are widely used to measure the amount of nucleic acid in a preparation. If the DNA sample is pure (i.e., without significant amounts of contaminants such as proteins, phenol, or other nucleic acids), spectrophotometer measurement of the amount of UV irradiation absorbed by the bases is simple and accurate. If the amount of DNA or RNA is very small or if the sample contains significant quantities of impurities, the amount of nucleic acid can be estimated from the intensity of fluorescence emitted by ethidium bromide or DNA fluorometer.

Electrophoresis through agarose gel is the standard method used to identify and separate DNA fragments. DNA molecules have a constant negative charge per unit length and they move towards positive charge (anode) under electrophoresis, thus aiding the separation of DNA according to their molecular weight.
