MEDOX-BIOTM SDS Page Teaching Kit

SDS PAGE is a widely employed technique for analyzing & resolving complex proteins and also for the determination of molecular weight of polypeptides with markers of known molecular weight. The polymerization of acrylamide & bis-acrylamide is initiated by adding freshly prepared APS to the mix followed by TEMED. The gel after electrophoresis is stained where the dye penetrates the entire gel. The dye only sticks permanently to the proteins and the 'Destaining' process washes out the excess dye leaving a clear protein band whose molecular weight can be determined by using markers. APS is used as catalyst for the polymerisation of polyacrylamide gel. The polymerisation reaction is driven by free radicals that are generated by an oxido-reduction reaction in which TEMED is used as adjunct catalyst.

MEDOX-BIOTM Silver Staining Teaching Kit

Silver staining is the most sensitive non-radioactive method of staining polypeptide with silver salts after separation of proteins by SDS-PAGE. In this case, the process relies on differential reduction of silver ions that are bound to the chain of amino acids. This process can detect polypeptide at nanogram level present in a single band.

MEDOX-BIOTM Protein Estimation Teaching Kit

The quantitation of protein in biological sample is of great importance for further research & development. Protein quantitation is widely utilized mostly for molecular biology research. This can be determined by using the following colorimetric methods.
Bradford Assay : This method is based on the principle that the protein binds to CBBG-250 in acid solution and forms a complex. The dye binds strongly to positively charged groups of proteins and also to hyrdophobic regions in proteins. As a result blue colour is formed.
Lowry Assay : In this method the proteins form copper protein complex and is reduced by the addition of Folin-Ciocalteu phenol reagent which gives a blue colour complex. The intensity of colour depends on the aromatic aminoacid content of a protein.
Biuret Assay : The Biuret is a good general protein assay for batches of material for which yield is not a problem. Under the alkaline condition, the copper ion is bound to nitrogen atoms of the peptide bonds in protein which in turn produces purple colour at a high temperature with a maximum absorption at 540-560nm.

ALP is a hydrolase enzyme responsible for removing phosphate group from many types of molecules including nucleotides, proteins and alkaloids. The enzyme is most effective in an alkaline environment. ALP is phosphomonoesterase with broad substrate specificity.

In this kit, assay of ALP makes uses of a chromogenic substrate called p-nitro phenyl phosphate. The product p-nitro phenol provides an easy quantitative way to monitor enzyme action.

Features & Application

* ALP has a useful tool in molecular biology laboratory, for removing the 5' phosphatase from plasmid DNA and vectors.
