Column chromatography is one of the most useful methods for the separation and purification of both solids and liquids when carrying out small-scale experiments. Column chromatography is another solid-liquid technique in which the two phases are a solid (stationary phase) and a liquid (moving phase). The theory of column chromatography is analogous to that of thin-layer chromatography. The most common adsorbents - silica gel and alumina - are the same ones used in TLC. The sample is dissolved in a small quantity of solvent (the eluent) and applied to the top of the column. The eluent, instead of rising by capillary action up a thin layer, flows down through the column filled with the adsorbent. Just as in TLC, there is an equilibrium established between the solute adsorbed on the silica gel or alumina and the eluting solvent flowing down through the column

MEDOX-BIOTM Gel Filtration Chromatography

Gel filtration chromatography is also called molecular exclusion or gel permeation chromatography. It separates the biomolecules based on their size. Gel filtration chromatography consists of porous beads with a well-defined range of pore sizes. A sample in a solvent is passed through the beads; molecules that are smaller than the holes in the beads get hung up in the beads. Therefore, the smaller molecules move through the column more slowly than larger molecules. Depending on the molecular weight, the protein may be separated, based on the size alone and fractions containing the isolated proteins can be collected.

MEDOX-BIO™ Purification Of Sequence Specific DNA Binding Protein Teaching Kit (By Affinity Chromotography)

Affinity purification of sequence-specific DNA binding proteins that is fast and effective. Complementary chemically synthesized oligodeoxynucleotides that contain a recognition site for a sequence-specific DNA binding protein are annealed and ligated to give oligomers. This DNA is then covalently coupled to Sepharose CL-2B with cyanogen bromide to yield the affinity resin. A partially purified protein fraction is combined with competitor DNA and subsequently passed through the DNA-Sepharose resin. The desired sequence-specific DNA binding protein is purified because it preferentially binds to the recognition sites in the affinity resin rather than to the nonspecific competitor DNA in solution. For example, a protein fraction that is enriched for transcription factor Sp1 can be further purified 500- to 1000-fold by two sequential affinity chromatography steps to give Sp1 of an estimated 90% homogeneity with 30% yield. In addition, the use of tandem affinity columns containing different protein binding sites allows the simultaneous purification of multiple DNA binding proteins from the same extract. This method provides a means for the purification of rare sequence-specific DNA binding proteins, such as Sp1 and CAAT-binding transcription factor.

Chromatography is used to separate organic compounds on the basis of their charge, size,
shape and their solubility. A chromatography consists of mobile phase and a stationary phase.
In Ion Exchange chromatography, separation is based on charge of the molecules. Proteins
contain many ionizable groups on the side chains of their amino acids as well as their aminoand
carboxyl- termini.
This technique is called anion exchange. If the beads are negatively charged, they bind to
positively charged molecules (cation exchange). Thus, a scientist picks the resin to use, based
on the properties of the protein of interest. During the chromatography, the protein binds to the
oppositely charged beads. Crude sample is loaded from top of equilibrated column and washed
with wash buffer to remove unwanted proteins. Once the contaminant protein is separated from
the protein of interest(i.e Lysozyme), a high salt buffer is used to elute the same from the
column. Then the lysozyme is assayed by using spectrophotometer and compared with
standards.

MEDOX-Bio® Immobolized Metal Ion Affinity Chromatography Teaching Kit

Immobilized metal ion affinity chromatography (IMAC) is based on the specific coordinate covalent bond of amino acids to metals, particularly histidine. This technique works by allowing proteins with an affinity for metal ions to be retained in a column containing immobilized metal ions, such as cobalt, nickel, copper for the purification of histidine containing proteins or peptides, iron, zinc or gallium for the purification of phosphorylated proteins or peptides. Many naturally occurring proteins do not have an affinity for metal ions, therefore recombinant DNA technology can be used to introduce such a protein tag into the relevant gene. Methods used to elute the protein of interest include changing the pH, or adding a competitive molecule, such as imidazole.
