Normally pH affects the growth of microorganism, the activity of enzyme. Especially those enzyme involved in biosynthesis and growth. Each species of microorganisms posses a definite pH range and a distinct growth medium. Acidophiles have a growth optimum between pH lead 5.5 Neutrophiles have between 8.5 to 11.5 majority of bacteria and alkalophiles have 8.5 to 11.5 majority of bacteria and Protozoa are Neutrophiles.Many bacteria produce metabolic acids that may lower the pH and inhibit the growth. To prevent the buffers are added to culture media to neutralize acids produced for e.g.: Peptone in complex media act as buffers. Phosphate salts are often added as buffers in chemically defined media.

Microbial growth is directly dependent on how temperature affects cellular enzymes. With increasing temperatures, enzyme activity increases until the three- dimensional configuration of these molecules is lost because of denaturation of this protein structure. On the other hand, as a temperature is lowered towards the freezing point, enzyme inactivation occurs and cellular metabolism gradually diminishes. All bacteria can be classified into one of three major groups, depending on their temperature requirements, Psychrophiles, Mesophiles, Thermophiles.

Bacteria are separated from environment by selectively permeable membrane. The membrane can be affected by changes in the osmotic pressure of the surrounding. Osmotic pressure is a force developed when a solution of different solute concentration are separated by the membrane that is permeable only to the solvent. This Phenomenon is known as plasma lysis cells become dehydrated and can't grow. In an isoelectric solution the concentration of solutes inside and outside the cell will be equal. Therefore bacteria placed in an isoelectric solution grows normally.

MEDOX-BIOTM Antibiotic Sensitivity Test Teaching Kit

Microorganisms are susceptible to variety of antibiotics. The sensitivity and mode of action of the antibiotic varies with microbial strains. Discs impregnated with known concentration of antibiotics are placed on a bacterial culture inoculated in agar plate, which has to be tested.
The anti-microbial agents diffuse through the agar and prevent the growth of microorganisms. The diameter of antibiotic zone is found to be directly proportional to the bacterial resistance.
