The formed elements of blood include erythrocytes, leukocytes and thrombocytes. Erythrocytes are non-nucleated middle sized cells (6.7-7.7 mm in diameter) and constitute the highest number of formed elements in the circulation. Leukocytes are nucleated and largest of all the cells with a wide range of size (9-20 mm in diameter). Leukocytes include granulocytes, lymphocytes, and monocytes. Granulocytes are further subdivided into neutrophils (multi lobed nucleus), esinophils (bilobed nucleus) and basophils (lobed nucleus). Thrombocytes are very small irregular bodies that play an important role in the coagulation of blood.

MEDOX-BIOTM Isolation Of Lymphocytes ateaching Kit

Primary source of lymphoid cells is the peripheral blood. Lymphocytes constitute 20%-40% of the body's WBC and 99% of the cells in the lymph. There are approximately 1010-1012 lymphocytes in the human body. Using Ficoll-conray density gradient centrifugation, the blood samples can be separated into different layers. This method involves the separation of cells based on their density using density gradient solution.

MEDOX-BIOTM Isolation Of Monocytes Teaching Kit

Heparinized whole blood is layered on top of a density gradient material (Ficoll/Hypaque) and subjected to a centrifugal force. Since erythrocytes (RBC) and PMN are denser than the gradient material, they penetrate the gradient material and sediment to the bottom of the centrifuge tube. The lighter mononuclear cells (lymphocytes and monocytes) sediment to the plasma-density gradient interface. At equilibrium, the mononuclear cells are carefully aspirated and saved.

The remaining plasma and gradient material are aspirated down to the RBC interface. The mononuclear cells are layered onto gelatin-coated flasks, allowing the monocytes to adhere. The non-adherent mononuclear cells (lymphocytes) are carefully decanted. The adherent monocytes are dislodged from the gelatin-coated surfaces with vigorous pipetting. The RBC cushion (which also contains PMN) is resuspended to the initial whole blood volume. Dextran 500 is added to a final concentration of 1% and mixed. The RBC is allowed to sediment to the bottom of the centrifuge tube. The PMN remain in suspension. The PMN-rich supernatant is carefully aspirated.

Six different types of WBC are normally found in the blood. They are polymorphonuclear neutrophils, polymorphonuclear esinophils, polymorph nuclear basophils, monocytes, lymphocytes and occasionally plasma cells. Blood transports WBCs from the site of origin to their site of action. WBCs can be detected in blood only during this passage. The lymphocytes have life spans of months as needed for these cells. The platelets in the blood are totally replaced approximately once every 10days.

The major function of RBC, also known as erythrocytes is to transport hemoglobin, which in turn carries oxygen from the lungs to the tissues. The red blood cells have other function besides simply transport of hemoglobin. The shape and size of RBC: Normal RBC is biconcave discs having a mean diameter of approximately 7.5mm and in the center of 1m or less. The average volume of the red blood cells in 83mm3 .
The blood is diluted and spread over an area of known dimensions on a clear slide in the form of thin film. The cells are counted and multiplied by the volume of blood and its diluting factors.

The immune system in higher organisms is composed of many lymphocytes subpopulations. In the human, one subpopulation of lymphocytes interacts in-vitro with unsensitized sheep erythrocytes to form structures termed "rosette". Rosette consists of a central lymphocyte surrounded by a cluster of attached sheep erythrocytes. The adherence of sheep red blood cells (SRBC) to a lymphocyte results in a formation termed a rosette. Rosette are formed by a small percentage of lymphocytes from peripheral blood of normal nonimmunized people. The significance of rosette formation in man is not well understood. Its role as an immuno logical tool is still controversial, although inhibition of the normal rosette formation by anti-lymphocyte serum (ALS) has been shown to be a very sensitive index of the immunosuppressive activity of various ALS preparations.
