Plasmids are small double stranded circular, extrachromosomal DNA molecules that are capable of replicating independently. They exist in multiple copies. The isolation of plasmid involves growth of bacterial culture, harvesting & lysis of the bacterial cell and the purification of the plasmid DNA. The obtained plasmid is resolved by using horizontal agarose gel electrophoresis.

MEDOX-BIOTM Genomic DNA Extraction Teaching Kit (ANIMAL)

DNA in the sample is released from blood cells using lysis buffer. The cell membrane of the animal tissue is weakened by EDTA in the Solution-I.SDS in Solution II denatures the cellular proteins. Phenol Chloroform removes all the protein contamination. The high molecular weight genomic DNA in aqueous phase is precipitated with ethanol or isopropanol (Solution-III) and the salts are removed by washing with 70% alcohol.

Features & Application
* Simple method of Genomic DNA isolation from animal tissue.
* DNA is of suffient purity for direct use in any application, e.g. PCR, Restriction digestion, cloning, and DNA sequencing, Southern blot analysis.

MEDOX-BIOTM Genomic DNA Extraction Teaching Kit (WHOLE BLOOD)

This kit is designed to isolate the genomic DNA from white blood cells. By using lysis solution red blood cells are lysed and white blood cells are left intact. The white blood cells and their nuclear membrane are lysed and solubilized by another solution. Then the proteins and other cellular components are removed by phenol-choloroform extraction. Finally the high molecular weight genomic DNA is precipitated by using absolute alcohol.

Features & Application
* The yield would be 5 - 10µg of DNA per ml of blood
* The extraction of genomic DNA from fresh blood by using solution based procedure

MEDOX-BIOTM Yeast Plasmid DNA Extraction Teaching Kit

A rapid technique for purifying plasmids from yeast Saccharomyces cerevisiae is described that yields high-quality DNA suitable for bacterial transformation, yeast transformation, and direct DNA sequencing. The method requires only small culture volumes and proprietary bacterial plasmid mini preparation kits that allow one to simultaneously prepare a large number of samples in a very short period of time while avoiding the use of toxic organic chemicals. Both yeast single-copy CEN/ARS and high-copy 2micro shuttle plasmid can be isolated using this method. This technique is useful for plasmid purification from yeast two-hybrid experiments as well as yeast genetics and molecular biology experiments.

MEDOX-BIOTM DNA Extraction Teaching Kit (PLANT)

The protocol consists of 2 steps. The rigid cell wall of the plant cell is weakened and lysed by osmotic shock. Solution II denatures the cellular proteins. Extraction with phenol/chloroform denatures the protein and removes SDS- Protein / polysaccharide complexes. The high molecular weight genomic DNA in aqueous phase is precipitated with 100% ethanol and further the salts are removed by washing with ethanol.

MEDOX-BIOTM Lambda Phage Isolation Teaching Kit

A scaled up lysate is used to prepare a large quantity of highly purified phage DNA. Host bacteria grown to A650-0.6 are infected with 105 -10 8 phage /ml. Following infection, the cells are shaken vigorously until cell lysis occurs. Remaining labile cells are lysed with chloroform and all debris is removed with a low speed spin. The phage is concentrated by PEG precipitation and purified by glycerol step gradient. The phage capsids are then degraded and the DNA is isolated.
