MEDOX-BIOTM DNA FingerPrinting Teaching Kit
(Using RAPD Technique)

DNA Fingerprinting by Random Amplified Polymorphic DNA method is used in Genetic analysis of organisms at molecular level. The primers approximately 10 bases in length base are used. Because the primer length is shorter, the temperature of the annealing reaction is reduced to less than 40°C.
RAPD is relatively insensitive to the integrity of the material. No blotting techniques are required. This decreases the amount of time required to obtain results.

Single nucleotide polymorphisms may fall within the coding sequences of genes, noncoding regions between genes. Genetic diseases and other related disorders are associated with SNP. MEDOX-BIOTM Long PCR Amplification Teaching Kit

Amplification of PCR products longer than 3-5kb is often compromised by non-specific primer annealing, sub optimal cycling conditions and secondary structure in the DNA template. Lengthy optimization is often necessary, varying factors such as cycling conditions, primers, and dNTP concentrations and special additives. The high rate of incorporation of incorrect basis by thermostable polymerases such as Taq that lack an editing function.
The incorporation of mismatched base at the 3' end of a growing strand may cause the enzyme to seize up and may limit the size of the PCR products. Proofreading DNA Polymerases, such as Pfu DNA poly usually contain inherent base pairs. PCR mixtures containing proofreading DNA polymerase consistently enable amplification of long products with high yield.

consuming optimization.

In vitro site-directed mutagenesis is an invaluable technique for studying protein structure-function relationships, gene expression and vector modification. Several methods have appeared in the literature, but many of these methods require single-stranded DNA as the template. The reason for this, historically, has been the need for separating the complementary strands to prevent reannealing.
Use of PCR in site-directed mutagenesis accomplishes strand separation by using a denaturing step to separate the complementing strands and allowing efficient polymerization of the PCR primers. PCR site-directed methods thus allow site-specific mutations to be incorporated in virtually any double-stranded plasmid; eliminating the need for M13-based vectors or single-stranded rescue
