In this immuno diffusion technique, a clarified agarose gel is used as a matrix for diffusion with precipitation. The antigen and antibody simply diffuse through the gel towards each other and precipitation results when the equivalence points have been reached on basis of antigenic determinant. Antigen antibody complex is formed in the agarose gel matrix and precipitation occurs at the point where the concentration of antigen antibody attain equivalence.

Immunoelctrophoresis is a combined technique of Immunodiffusion and Immuoectrophoresis. This process involves electrophoresis of antigen to separate its components in the agarose gel based on their charges, followed by diffusion of antigen with antibody loaded in the trough which diffuses and binds with the specific antigenic components and forms white precipitin arc or line.

MEDOX-BIOTM Single Radial Immunodiffusion Teaching Kit

The Single Radial Immunodiffusion is a simple but comparatively more sensitive method than any other Immunodiffusion technique. This technique is done mainly by incorporating the antiserum to a thin layer of agarose and then making the antigen to diffuse radicals and react with the constant amount of immobilized antibody and form the precipitin ring at their point of equivalence in proportion with antigen concentration. In this technique, antigen are allowed to diffuse from a well into an antibody containing gel. The precipitin rings are formed in the gel around the well.

This Counter current Immunoelectrophoresis kit is mainly based on the principle that, the antigen and antibody placed in wells are driven electrically to migrate faster towards each other.
