In simple staining, the bacterial smear is stained with a single reagent. Basic stains with a positively charged chromogen are preferred because bacterial nucleic acids and certain cell wall components carry a negative charge that strongly attracts and binds to the cationic chromogen. The purpose of simple staining is to elucidate the morphology and arrangements of bacterial cells. The commonly used basic stains are Methelene Blue, Crystal Violet, and Carbol Fuchsin.

MEDOX-BIOTM Gram Staining Teaching Kit

Differential staining requires the use of three chemical reagents that are applied sequentially to a heatfixed smear. The first reagent is called primary stain. Usually crystal violet (Hucker's) is used, as Primary stain, its function is to impart its colour to all cells. Gram's Iodine serves as a mordant, a substance that increases the cells affinity for a stain. In order to establish colour contrast, the second reagent (95% Ethyl alcohol) is used as decolorizing reagent. Safranin is used as a Counter stain. Gram Negative cells undergo decolorization, and absorb the counterstain. Gram- Positive cells retain the purple colour of the primary stain.

MEDOX-BIOTM Capsular Staining Teaching Kit

Capsule staining is more difficult then other types of differential staining procedures because the capsular materials are water soluble and may be dislodged and removed with vigorous washing. Capsule is a gelatinous outer layer that is secreted by the cell surrounds and adheres to cell wall.

The capsule stain uses two reagents. Crystal violet is used as the primary stain. At this point the cell and the capsular material will appear in dark colour. Copper sulphate is used as the decolorizing agent and as the counter stain. Since the capsule is water soluble, copper sulphate, rather than water is used to wash the purple primary stain out of the capsular material without removing the stain that is bound to the cell wall. At the same time, it acts as a counter stain as it is absorbed into the decolorized into the capsular material. The capsule will now appear light blue in contrast to the deep purple colour of the cell.

MEDOX-BIOTM Flagellery Staining Teeaching Kit

The flagella are untangled by adding 1% peptone broth to the overnight grown agar slant culture. The cells are then flooded with a mordant, which increases the stains affinity for the specimen. As the alcohol from basic Fuchsin (Primary stain used for flagella stain) evaporates, the dye Precipitates, and become adsorbed on the flagella, capsules or any other material which may be on the slide. Tannic acid blocks those sites that normally would be contrasted by the counter stain. (Zeihls carbol fuchsin). The cells are then washed with water and flooded with strong counter stain, which stains the other parts of the organism.

* This staining technique is simple enough to be employed in routine pure culture study in the place of the conventional test for motility.

MEDOX-BIOTM Acid Fast Staining Teaching Kit

Certain members of bacterial organisms particularly, Mycobacterium are resistant to other staining methods and can be visualized by the Acid fast staining. The characteristic difference between Mycobacteria and other organisms is the presence of a thick, waxy (lipoidal) wall that makes penetration by stains extremely difficult. Once the stain has penetrated, however, it cannot be readily removed even with the vigorous use of acid alcohol as a decolorizing agent. Because of these property, these organisms are called Acid-fast, while all other microorganism, which are easily decolorized by acid- alcohol, are non-acid- fast.

Mounting Lacto phenol cotton blue is a stain used for making semi permanent microscopic preparation of fungi. It Stains the fungal cytoplasm and provides a light blue background, against which the walls of hyphae can readily be seen. It contains four constituents, phenol which serves as a fungicide, lactic acid which act as a cleaning agent, cotton blue which stains the outer wall of the fungus and glycerin which gives a semi permanent preparation. A Permanent preparation may be made by incorporating polyvinyl alcohol in place of glycerin is to the medium.

MEDOX-BIO® Endospore Staining Teaching Kit

 

Vegetative cells are bacteria that are actively growing, metabolizing and dividing. When vegetative cells are subjected to environmental stresses such as nutrient deprivation they eventually die. However, some bacteria such as the Bacillus species and the Clostridium species can circumvent the problems associated with environmental stress by forming endospores. Because of the chemical composition of spore layers, the spore is resistant to the deleterious effect of excessive heat, freezing, radiation, desiccation, and chemical agents, as well as the commonly employed microbiological stains. The endospore can be stained by using strong penetraters like malachite green. After staining, spore will be green in colour and its surrounding will be seen in pink.

Simple staining is also referred as monochrome stains, since only one dye is employed for the coloration of bacterial smear. The surface of a bacterial cell has an overall acidic characteristic because of large amount of carboxyl groups located on the cell surface due to acidic amino acids. Therefore, when ionization of carboxyl groups takes place it imparts negative charge to the cell surface as per the following equation.

MEDOX-BIO® Metachromatic Granules Staining Teaching Kit

Metachromatic granules (volutin) are a type of cytoplasm inclusions found in many bacteria as well as in some fungi, algae and protozoa. The granules are composed mainly of polyphosphates, RNA and proteins. Although, speculations have been made as (i) reserve source of food, (ii) phosphate storage structure, (iii) link with energy metabolism and (iv) involvement in nucleic acid synthesis and cell division. The granules are found to be most prominent in old cultures, before starvation occurs. During this prominent looking stage the culture has been stained and viewed.
